AUTOMATED SAMPLE PREP

The Laminar Wash™ technology comprises a wall-less plate and laminar-flow fluidics that enables washing of suspension cells at a fraction of the hands-on time while yielding consitently higher cell retention and viability. Using a total of 192 individual nozzles, the Laminar Wash™ instrument gently flows buffer across the top of settled cells to remove debris and unbound antibodies instead of subjecting cells to the significant stress associated with traditional centrifugation methods. The result is reliable, consistent, and superior data in flow cytometry, single-cell sequencing, or other downstream applications.

Automation options abound with Laminar Wash systems. The HT2000 system can be easily integrated into virtually any robotic arm or personal pipettor workstation using the Automation License Key, which includes an API. A fast and simple automated workflow can wash 10 plates in just over 20 minutes. The AUTO1000 represents a fully turnkey system that can perform end-to-end flow cytometry sample preparation, right out of the box, without any scripting. These systems represent elegant, fast, and low-cost alternatives to centrifuge automation, which involves many more complicated movements.

Besides the undue stress that centrifuges impart on cells during washing steps, they also require several complicated steps that are difficult to automate. Centrifugation of suspension cells is a roadblock to otherwise straightforward automation.

Reproducibility of Low Numbers of Cells

Laminar Wash consistently yields higher cell retention compared to the traditional centrifuge, even at low cell numbers. Presented are isolated naive T-cells from freshly-thawed PBMCs, at starting cell numbers of 10k, 50k, and 100k. Actual cell counts of CD3+ and %CD3+ in singlets demonstrate consistently higher cell retention rates compared to equal numbers of washes.

Increased Reproducibility of Data from Critical Samples

Laminar Wash technology demonstrates greater reproducibility between replicates compared to traditional centrifugation. Presented here is a flow cytometric crossmatch assay (FXCM), widely used in human leukocyte antigen (HLA) labs to predict donor-recipient compatibility. Using three test sera and two controls in eight replicates, Laminar Wash produced a lower coefficient of variance in B-cell median channel fluorescence (MCF) values of 7.9%, versus 12.4% in the centrifugation method. Higher MCF values on the positive control also gave greater FCXM sensitivity, which is highly valued in clinical settings.
