A quicker, simpler path to better data

With the HT2000’s laminar washing technology, cells are deposited onto a 96-well laminar wash plate, where a gentle flow of buffer washes away unwanted debris, such as unbound antibody or dye. Unlike with centrifugation, very little stress is placed upon the cells, ensuring the integrity of cell surface markers. The thoroughness of the laminar washing process produces cleaner, better segregated data than centrifugation-based wash methods and the gentleness of the technique enables high cell retention and viability.

Compared to a traditional, centrifuge-driven cell washing workflow, laminar cell washing is simpler and faster. With the HT2000, you can get complete washing of 96 samples in just 3 minutes.

How our plate and nozzle design wahes your cells

The power of Curiox’s laminar cell washing technology lies in the unique design of the 96-sample plate. Each plate contains an array of hydrophilic spots that suspension cells can rest on via gravity. The material surrounding each spot is highly hydrophobic, acting as an impenetrable wall that separates your samples.

Upon entry into the instrument, two nozzles descend onto each side of every spot, one to dispense wash buffer and the other to aspirate the buffer and debris away.

Each spot can wash as many as 10 million cells down to a single cell without the mechanical stress and cell losses associated with centrifugation.

See how dye is washed away from green fluorescent polystyrene beads during 2 minutes of 7 aspiration and dispense wash cycles. At the end of the cycle, the dye is gone but the polystyrene beads remain in the spot.

Laminar wash advances with neutrophils and monocytes retention for improved differential blood count
With 100ul of human whole blood, a comparison study using a 16-color flow cytometry panel focused on T cells was conducted by Laminar Washing. Whole blood lysed with ammonium chloride buffer was first centrifuged once to reduce volume, and then aliquoted to incubate with antibodies and washed on either Laminar Wash or centrifuge in polystyrene tubes. There is clear resolution of rare T cell populations such as MAIT, gamma-delta T cells and regulatory T cells with replacement of centrifuge with Laminar Wash in one wash step. The elevated retention for neutrophils and monocytes was observed and quantified via direct acquisition onto a BD Fortessa, allowing minimal cell perturbations.
